The highly skilled Federale Machete (Danny Trejo) is left for dead after clashing with notorious Mexican drug-dealer Torrez (Steven Seagal). Three years later, in Texas, the dangerous businessman Booth (Jeff Fahey) offers Machete the opportunity to make $150,000 by assassinating the unscrupulous right-winged Senator McLaughlin (Robert De Niro). The latter is running for reelection on an anti-immigration platform. The assassination is a set-up designed to make McLaughlin look like an injured martyr.

Machete escapes again and sets out to settle scores both old and new. He gets some help from Luz (Michelle Rodriguez) the head of The Network, an organization that helps Mexican immigrants cross the border. Sartana (Jessica Alba) is an immigrations officer torn between enforcing the law and doing what is right. Further there's Machete's brother Padre Cortez (Cheech Marin) who's an old-school guerrilla and also a Catholic priest. With numerous other Mexican allies, Machete seems destined to lead the war against the political system that forbids Latinos to work in the USA.
